abstract | The present invention uses carboxypeptidase E (CPE) genes that are known to be involved in fat metabolism, such as obesity and body fat accumulation in mammals, from Korean native cattle, a native animal gene source using genetic engineering and gene genome analysis technology. It contains information on the development of DNA markers of carboxypeptidase E (CPE) gene, which significantly affects marbling and muscle fat thickness.n n n In the present invention, the genes of the Intron 4 region and the Exon 5 region of the Hanwoo carboxypeptidase E (CPE) gene were cloned and analyzed by sequencing to analyze the gene structure. Single nucleotide polymorphisms (SNPs) were identified and their associations with meat and carcass traits of Hanwoo.n n n Thymine (T) ↔ at 445th in intron 4 region and 601th in exon 5 region, respectively, in the 511 bp sequence including the intron 4 and exon 5 regions of the Hanwoo carboxypeptidase E (CPE) gene A single nucleotide polymorphism (SNP) site caused by cytosine (C) base substitution was found. These monobasic polynucleotide sequences were digested with NlaIII and DdeⅠ restriction enzymes using PCR-RFLP (restriction fragment length polymorphism) techniques, respectively, to determine three genotypes (T / T, T / C and C / C). Was detected. Among them, a new finding was found that the T / C genotype was significantly related to intramuscular fat and back fat thickness of Hanwoo.n n n Accordingly, the present invention is directed to a conventional selection method for selecting specific SNP genotypes of the carboxypeptidase E (CPE) gene, which has a significant effect on the marbling and backfat thickness of Korean cattle.
