abstract Methods for administering mitomycin C to multi-drug resistant cells and methods for reducing the toxicity of mitomycin C are described. In this method, mitomycin C is provided in the form of a prodrug conjugate wherein the drug is bound to a hydrophobic site, such as a lipid, via a cleavable dithiobenzyl bond. Dithiobenzyl bonds are susceptible to cleavage by mild thiolysis, allowing mitomycin C to be released in their native form. The bond is stable under non-reducing conditions. Prodrug conjugates may be incorporated into liposomes for administration in in vivo reducing conditions or in response to administration of exogenous reducing agents.
