abstract The present invention provides methods and compositions useful for modulating signaling through toll-like receptors. The method of the present invention comprises contacting a TLR-expressing cell with a small molecule having a core structure comprising two or more rings. Particular compounds are 4-primary amino quinoline. Many of these compounds and methods are particularly useful for inhibiting immune stimulation associated with one or more of TLR9, TLR8, TLR7 and TLR3. The method can be used to treat autoimmunity, inflammation, allergy, asthma, graft rejection, graft versus host disease, infection, sepsis, cancer and immunodeficiency.
