abstract |
Methods of synthesizing polymer conjugates of cytokines and their receptor-binding antagonists, in particular unglycosylated interferon-β, are provided which maintain very high biological efficacy. Preparation of the polymer conjugates according to the method of the present invention reduces the steric inhibition of receptor-ligand interactions generally caused as a result of the adhesion of polymers to receptor-binding regions of cytokines and their agonism and antagonist analogs. Or prevent it. The invention also provides conjugates and compositions prepared by such methods. The conjugates of the present invention maintain a higher level of biological efficacy than those produced by conventional polymer coupling methods that are not intended to avoid receptor-binding regions of cytokines. In in vitro assays, the biological efficacy of the conjugates of the nonglycosylated interferon-β of the present invention is substantially higher than that of the unconjugated interferon-β and is similar to that of the glycosylated interferon-β 1a. The conjugates of the invention also exhibit prolonged half-life in vivo when compared to the corresponding unconjugated cytokine.
