abstract |
The present invention relates to methods of genotyping single nucleotide differences in nucleic acid samples. More specifically, the present invention provides a method for identifying nucleotides of a polymorphic site or a group of polymorphic sites in a genomic DNA sample. The method utilizes tag-labeled primer extension in which a set of tag sequences corresponds to the identity of the nucleotide at the polymorphic site. Primer extension products are PCR amplified using a common set of tag-specific primers, downstream primers with identifiable labels. After separation by size and / or charge, detection of the identifiable label in the product of expected size determines the identity of the nucleotide at the polymorphic site. The method is well suited for genotyping multiple single-nucleotide differences in a series of reactions. |
