abstract |
The present invention provides improved chimeric glycoproteins (GPs) and improved lentiviral vectors pseudotyped with the glycoproteins. The invention also provides methods and compositions for the preparation of said glycoproteins and vectors. The improved chimeric GP encodes the transmembrane extracellular domain of GALV or RDl14 GP that is fused to the cytoplasmic tail of MLV-A GP. Vectors pseudotyped with the GALV / TR and RD114 / TR GP chimeras have a significantly higher threshold than vectors coated with parental GP. Furthermore, the RDl14 / TR- pseudotyped vector is efficiently concentrated and resistant to inactivation caused by enrichment of human and macaque monkey serum. RDl14 GP-pseudotyped lentiviral vectors are particularly useful for in vivo gene delivery applications. |
