abstract | Pulmonary delivery by aerosol inhalation has received a lot of attention as an attractive alternative to intravenous, intramuscular, and subcutaneous injection because this approach eliminates the need for syringes and needles for injection. The present invention provides cellulose-based dry powder filled capsules that are particularly useful for use with dry powder inhalers. These capsules not only function well in combination with conventional DPI, but also in combination with conventional powder filling techniques, saving time, labor and cost. The present invention further provides a novel method of theoretically determining appropriate optimum conditions for the preparation of such powder filled capsules. Specifically, it is important that the dry powder formulations for long term storage do not exceed the critical moisture point, which is the point at which the powder loses physical and chemical stability. The present invention describes a means of predicting equilibrium moisture content, which in turn can be used to establish a suitable capsule preparation and filling protocol. |
