abstract | In a sample obtained at a site other than the tumor primary of the patient, a first primer homologous to a portion of a specific sequence in RNA derived from tumor cells and a second primer complementary to a portion of the specific sequence (one of which is 5 'end) (C) synthesizing cDNA by enzymatic action having RNA dependent DNA polymerase activity using (1) the specific sequence as a template, and (2) An enzyme having DNA-dependent DNA polymerase activity by digesting RNA strands in RNA-DNA double strands by the enzyme having ribonuclease H activity (generation of single stranded DNA), and (3) using the single stranded DNA as a template. Action to form double-stranded DNA having a promoter sequence capable of being transcribed into RNA complementary or homologous to the particular sequence, and (4) inhibiting RNA polymerase activity. That is how to transfer the double-stranded DNA by enzymatic action to (is used as a template in the cDNA synthesis after in the above reaction (1)) RNA transcription product, and detecting the tumor cells micrometastasis, comprising detecting the mRNA. |
