abstract | The present invention relates to combination therapies for treating cell proliferative disorders associated with methylthioadenosine phosphorylase ("MTAP") deficient cells in a mammal. The combination therapy selectively kills MTAP deficient cells by administering an inhibitor of de novoinosinate synthesis in combination with an antitoxin. Wherein the inhibitor of de novo inosinate synthesis is an inhibitor of glycineamide ribonucleotide formyltransferase ("GARFT") and / or aminoimidazolecarboximide ribonucleotide formyltransferase ("AICARFT"), and the antitoxin is an MTAP substrate (Eg, methylthioadenosine or “MTA”), an MTA precursor, an analog of the MTA precursor, or a precursor of an MTAP substrate. |
