abstract | The present invention provides methods and compositions for integrating a nucleotide sequence of interest into the genome. In particular, the methods of the present invention comprise contacting a lentiviral vector with an oocyte, early stage embryo or blastocyst plasma membrane under conditions such that a viral vector can be introduced to incorporate the desired nucleotide sequence into the genome. The method further comprises culturing the oocytes or embryos in contact with the lentiviral vector under conditions capable of forming preimplantation embryos. The preimplantation embryo can then be delivered to the receptor vertebrate, in which case the embryo can be generated from one or more genetically modified animals. Thus, the methods and compositions of the present invention can produce genetically modified animals, specifically vertebrates and more specifically mammals. |
