The present invention provides an accurate folded biologically inert monomer, oligomeric or polymeric biologically active granular cell affinity chromatography using immobilized metal affinity chromatography from granulocyte colony stimulating factor (G-CSF) and bound G-CSF molecules. A method for isolating G-CSF, which enables the separation of active monomeric G-CSF molecules. The process of the present invention may optionally be carried out advantageously under natural conditions, if desired. This gives a biologically active G-CSF with a purity greater than 95%. Only two further chromatography steps, namely cation exchange chromatography and gel filtration, are preferably applied to remove trace impurities. The whole method allows for higher yields of G-CSF with greater than 99% purity. The described method is particularly suitable for the industrial production of G-CSF. |
