abstract | The present invention relates to a high sensitivity assay using a method for gene-specific priming and amplification of mRNA derived from rare cells and rare transcripts found in blood. This assay can detect rare mRNAs (10 copies / cell) derived from 1-10 cells isolated through immunomagnetic enrichment. This assay is an improvement over complex PCR, which enables efficient detection of rare coding sequences for circulating carcinoma cells in the blood.n n n The methods of the invention are useful for profiling cells isolated from tissues and body fluids, and also assist in the clinical diagnosis of various carcinomas, including early stage detection and classification of circulating tumor cells. By monitoring the nucleic acid and protein profiles of the cells in conventional or microarray formats, it is possible to facilitate the regulation of therapeutic intervention, including delaying treatment, monitoring response to treatment, confirming recurrence, and determining whether it is armored. |
