abstract | The present invention is directed to a method of preventing the modification of a synthetic oligonucleotide or oligonucleotide analogue while removing one or more β-cyanoethyl groups from the oligonucleotide or oligonucleotide analogue. The method comprises contacting an oligonucleotide or oligonucleotide analogue with a basic solution with one or more acrylonitrile conservatives such as t-butylamine for a sufficient time at a temperature sufficient to remove one or more β-cyanoethyl protecting groups. . The invention also relates to methods of making synthetic oligonucleotides or oligonucleotide analogs. |
