abstract Cyclic oxyguanidine compounds or solvates, hydrates or pharmaceutically acceptable salts thereof are disclosed, including compounds of formulas (I and II) that inhibit proteolytic enzymes such as thrombin (wherein R 1 , R 3 -R 6 , R 21 -R 26 , L, Y, Z and A are disclosed herein). Also provided are methods of preparing compounds of formula (I and II). The novel compounds of the invention are inhibitors of potent proteases (particularly serine proteases of the trypsin type, for example chymotrypsin, trypsin, thrombin, plasmin and factor Xa). Certain compounds are useful intermediates that produce antithrombotic activity through direct and selective thrombin inhibition, or to prepare compounds with antithrombotic activity. The present invention relates to a pharmaceutically acceptable carrier which inhibits the loss of blood platelets, inhibits the formation of blood platelet aggregates, inhibits the formation of fibrin, inhibits thrombus formation, and inhibits embolism in mammals. Compositions comprising the compounds of the invention. Other uses are embedded in or physically embedded in devices used for blood collection, blood circulation and blood storage, such as catheters, hemodialysis, syringes and tubes for blood collection, and materials used in the manufacture of blood lines and stents. It is used as an anticoagulant.
