abstract | The present invention relates to a method for producing enkephalin using proteolytic enzymes in a single system of water-mixed organic solvents. The trileu is a leucine-enkephalin synthetic precursor using chymotrypsin and thermolysin through proteolytic reverse reaction in non-aqueous solvents. After synthesizing the peptide and the dipeptide, respectively, the method of producing pentapeptide enkephalin using papain from the two substrates has much higher stereospecificity and position specificity and a significantly lower need for protecting group than conventional chemical synthesis. In addition, it has an excellent effect on the active enkephalin synthesis, such as no by-products and no problem of racemate production. |
