abstract The present invention is an inhibitory peptide capable of inhibiting β-wrinkle sheet formation in amyloid β-peptide. The inhibitory peptides are β-sheet breaker peptide homologs designed by chemically modifying β-sheet breaker peptides capable of inhibiting β-wrinkled sheet formation in amyloid β-peptides. The present invention also includes inhibitory peptides capable of inhibiting conformational changes of prion PrP proteins associated with amyloidosis. The inhibitory peptide is a β-sheet breaker peptide homolog designed by chemically modifying the β-sheet breaker peptide capable of inhibiting the conformational change of prion PrP protein associated with amyloidosis. The present invention also encompasses peptide analogs having a PMiAβ5 structure. In another embodiment, the peptide analog has a PMiPrP13 structure. In another embodiment, the peptide analog has a PMiPrP5 structure of the formula
