The present invention provides an antibody comprising (a) a DNA comprising a full length copy of a genomic ran (gRNA) or RNA virus; Or (b) DNA comprising one or several fragments of an RNA viral gRNA, encoding one or more structural or nonstructural protein and RNA dependent RNA polymerase; Or (c) cloning a DNA having at least 60% homology with the sequence of (a) or (b) into a bacterial artificial chromosome (BAC). Also provided are DNA comprising RNA dependent RNA polymerase and sequences derived from genomic RNA (gRNA) of coronaviruses having at least 60% sequence homology with the native sequence of the virus encoding one or more structural or nonstructural proteins. A fragment of this DNA can be transcribed into an RNA dependent RNA polymerase that can be assembled into a virion. Also disclosed are the use of these nucleic acids in the preparation of RNA or virions and agents comprising these DNA, viral RNAs or virions. |
