abstract | The present invention provides a method of separating said nucleic acid molecule from a solid support to which said nucleic acid molecule is attached, comprising selectively cleaving a nucleic acid molecule at said non-standard nucleotide site, wherein said non-standard nucleotide is incorporated at a predetermined site within said molecule. do. The nucleic acid molecule may be a chimeric molecule comprising a nucleic acid (nucleotide sequence) and other molecular components of different chemical properties. In addition, this aspect of the invention provides a chimeric molecule (or structure) comprising a nucleotide linker sequence comprising, at a predetermined site, a selectively cleavable non-standard nucleotide bound to a functional group that may be an affinity linker or reporter group. The present invention has utility in a variety of uses, for example in molecular biology methods such as nucleic acid synthesis, amplification or sequencing, nucleic acid isolation or in isolation or analysis methods based on affinity. |
