titleOfInvention Coating of solid surfaces with activated polyhydroxypolymers
abstract The present invention provides a simple and novel method for producing a solid surface coated with a water soluble activated polyhydroxypolymer. The method comprises contacting and rinsing a solid surface substantially free of amine, imino, or thiol groups with a coating solution comprising the activated polyhydroxypolymer to bond the activated polyhydroxypolymer to the solid surface. Optionally drying the solid surface. The contact reaction should take place in an aqueous medium having a pH of 1.5-10 or an ionic strength of 0.1-8. Preferred polyhydroxypolymers are polysaccharides, in particular dextran, and other natural polymers free of aldehyde groups such as cellulose, agarose and starch. Also preferred are synthetic polymers, in particular polyvinyl alcohol. Preferably the water solubility of the polymer is at least 10 mg / ml and its molecular weight is at least 1000. The activation of the polymer is preferably a functional group selected from tresyl, maleimido, cyanozen bromide, tosyl, trityl, pentafluorobenzenesulfonyl and vinylsulfone groups. Particularly preferred activated polyhydroxypolymers are tresylated, tosylated, or maleimated dextran. The solid surface is preferably a surface of an organic polymer (such as polystyrene), glass, ceramic or metal. The present invention also provides a solid surface obtainable by the method and the use of the surface for immobilizing biomolecules.
