abstract | The present invention relates to modified pneumolysin polypeptides that retain the immunogenicity of numericin while being so small or reduced that no hemolytic activity is detected compared to native numericin. The present invention also relates to methods of generating new numericin variants with these desired properties. In addition, the present invention provides immunocompositions that are useful as pharmaceutical compositions, including vaccines in which nontoxic, modified pneumolysin stimulates prophylactic immunity against Streptococcus pneumoniae. The vaccine is a composition in which modified pneumolysin is combined with a bacterial polysaccharide or contained in a weakened viral vector. In addition, the present invention also uses a non-toxic, modified pneumolysin toxin to stimulate an antibody against Streptococcus pneumoniae in a treated subject, which is then isolated and delivered to a secondary subject, thereby eventually streptococci to the secondary subject. It also relates to methods of providing Cous pneumoniae prophylaxis. |
