abstract |
The present invention comprises the steps of (a) collecting the fertilized egg or bovine somatic cells obtained by in vitro fertilization and in vitro culture of the cow and stored under washing and dispensing conditions; (b) isolating and preserving cell lines from various tissues of the bovine body; (c) dissecting a portion of the zona pellucida of the mature egg prepared in step (a) and denuclearizing 10-30% of the cytoplasm; (d) transplanting the donor cells prepared in step (b) into an egg whose nucleus has been denuclearized in step (c); (e) electrofusion of the cells transplanted and adhered in step (d) with a voltage of DC condition; (f) activating the fused cells in medium; And (g) post-activating and inspecting the activated cells, and then culturing the cells in a micro medium. In the step of the voltage under the DC condition, it is characterized in that the electrofusion is performed by adjusting the number of times of recovery x time to be within 60 s in total once or twice at 0.75 to 2.25 kv / cm. |
