abstract | A selection method for selecting one or more selectable genetically transformed cells from a cell population is disclosed. In this method, the cell population consists of selectable genetically transformed cells and possibly untransformed cells. Each selectable genetically transformed cell comprises a first nucleotide sequence that can be expressed and a second nucleotide sequence that is optionally expressed. In this method, when components or metabolic derivatives thereof are present in the medium at low concentrations, they are nutrients to both selectable genetically and non-transformed cells.n n n In this method, non-transformed cells are toxic when components or metabolites in the medium are present in high concentrations. The first base sequence encodes a gene product that, when present in high concentrations of a constituent or metabolic derivative therefrom, can convert it into nutrients for selectable genetically transformed cells. The method comprises the step of introducing a cell population into the medium, wherein the medium optionally comprises a high concentration of components or metabolites thereof. In this method the component metabolic derivatives are a source of carbohydrates and nitrogen for selectable genetically transformed cells. Alternatively, if some of the components in the method serve as metabolic substrates and are converted to induction substrates, the induction substrate can provide an allosteric ellect to the metabolite. |
