abstract | The present invention provides a method of analyzing a target nucleic acid, the method providing a binding probe array having a known sequence fixed to a substrate, wherein a portion of the bound probe is complementary to the first portion of the plurality of target nucleic acids; Providing a labeled probe of a known sequence, wherein a portion of the bound probe is complementary to the first portion of the plurality of target nucleic acids, and the particular second portion is adjacent to the particular first portion; Providing a material to be placed between the binding probe and the labeled probe to prevent probe mixing between the bound probe array and the labeled probe array; Adding a target nucleic acid to a labeled probe; Removing the material between the bound probe and the labeled probe and mixing the labeled probe with the bound nucleic acid together with the target nucleic acid under conditions that can identify that the complete probe is from one base mismatch; Linking coupled and labeled probes hybridized to a site adjacent to the target nucleic acid; Detecting a labeled probe connected to the coupled probe array. |
