abstract | The present invention relates to an enzymatic method for producing 7β- (4-carboxybutanamide) sephalosporonic acid using an altered enzyme D-amino acid oxidase of Trigonopsis variabilis produced in Escherichia coli. The method of expressing such an enzyme comprises (I) isolating DNA corresponding to a gene encoding enzyme D-amino acid oxidase; (II) removing the intron contained in the gene; (III) inserting the DNA fragment obtained above into a plasmid fragment that can be replicated in E. coli; (IV) fusing to the 5 ′ end of the structural region of the gene a synthetic assembler containing a nucleotide sequence encoding six histidines; (V) transforming the E. coli strain with the resulting recombinant plasmid; (VI) culturing the transformed cells of E. coli; (Iii) recovering the enzyme D-amino acid oxidase by affinity chromatography. |
