abstract The present invention relates to hot start PCR reaction mixtures comprising pyrophosphate and pyrophosphatase and to PCR methods for inhibiting non-specific amplification and primer-dimer formation occurring during PCR reactions. The hot start PCR reaction mixture of the present invention is prepared by adding pyrophosphate and pyrophosphatase to a PCR mixture containing a reaction buffer, MgCl 2 , four dNTPs, and a DNA polymerase. At this time, the electric pyrophosphate may be used by including 0.3mM to 5mM, preferably 0.4mM to 3mM, most preferably 0.5mM in the PCR mixture, and the electric pyrophosphatase is 0.0001U to 0.01U per 20 µl in the PCR mixture, Preferably from 0.0002U to 0.005U, most preferably 0.0003U is used. When the hot start PCR reaction mixture containing the pyrophosphate and the pyrophosphatase of the present invention is used, the production of non-specific amplification products is suppressed so that specific amplification products are effectively produced. It can be usefully used for amplification using or for suppressing non-specific amplification products generated during multiple PCR reactions.
