abstract | The present invention is an isoquinoline alkaloid compound extracted from Berberis vulgaris L., which not only promotes DNA cleavage mediated by topoisomerase II but also catalyzes topoisomerase II. It relates to a method of inhibiting the proliferation of cancer cells using berberubin to inhibit the activity. Berberubin is a reversible topoisomerase II-mediated DNA cleavage / reconnection reaction that traps topoisomerase II by trapping nicked circular DNA that is an intermediate between linear and circular DNA. Inhibition of DNA recombination, and directly inhibits the enzyme activity of topoisomerase even in the step before formation of the topoisomerase II-DNA cleavage complex. Thus, Berberubin is of value as a new anticancer agent targeting topoisomerase II. In addition, since berrubin has a mechanism of action different from that of the conventional topoisomerase II poison, it is expected to overcome drug resistance due to repeated use of conventional anticancer drugs. |
