abstract |
The present invention relates to an E. coli transformed with an expression vector into which an hemA gene encoding ALA synthase of Bradyrhizobium japonicum , one of ALA-producing strains, is inserted using a genetic recombination technique. It relates to a biosynthetic ALA manufacturing method using the same. The present invention constructs E. coli transformed with an expression vector into which hemA gene of Bradyrhizobium japonicum is inserted, and adds levulinic acid while culturing E. coli in a culture solution to which glycine and succinic acid are added. Provided is an efficient biosynthetic ALA preparation method for obtaining ALA. According to the biosynthetic ALA production method of the present invention, it takes a long time to produce ALA with the conventional ALA production method by other strains, culture conditions are difficult, feed-back inhibition of intracellular ALA dehydratase (feed ABA dehydratase) By improving the problems such as yield reduction caused by the (back-back inhibition) phenomenon, to produce a biosynthetic ALA that can be utilized as a herbicide without the purification step by efficiently producing ALA at a high yield of 3 to 5mM or more that can exhibit herbicidal activity can do. |
