abstract |
The present invention relates to methods and compositions for the treatment and restoration of cartilage or bone defects (defects in the total thickness of the joints) of humans and other animals. To induce cartilage formation, the cartilage depleting cells are filled with intracartilage defects in a matrix with pores of a size sufficient to survive in the matrix. The matrix contains antiangiogenic agents that act as functional blockers to prevent angiogenesis and bone growth into the cartilage region. The matrix filling the defects of the cartilage may also contain proliferative and chemotactic inducers and conversion factors present in suitable input systems. Functional blockers between bone and cartilage areas of the total thickness defect can be created by heat treating the bleeding area to form a temporary tissue blocker that prevents blood vessels and related cells from penetrating from the bone area to the cartilage area. If desired, the bone portion of the total thickness defect can be filled with a matrix with pores large enough to allow cells to increase the cell density of the matrix and form blood vessels. The matrix filling bone defects may include angiogenic and osteogenic factors present in a suitable input system. The methods and compositions of the present invention can assist bone and connective tissue regeneration for dental and other uses. |
