abstract | The present invention provides a method for screening an adapter protein gene that binds to the SH2 region of tyrosine kinase and SH2 of p56 lck that is involved in a signaling pathway that regulates T-cell activation with Src family tyrosine kinase selected using the same. Novel adapter protein genes that bind to regions and amino acid sequences inferred therefrom. We constructed a yeast transformed with a vector expressing a fusion protein between the activated kinase region of mouse p56 lck and the SH2 region and the DNA binding region of yeast GAL4, and using mouse T- From the cellular cDNA library, a gene expressing an adapter protein that binds to the SH2 region of p56 lck , that is, Lad (p56 lck- binding adapter protein), was selected, and the base sequence of the gene and the amino acid sequence inferred therefrom were analyzed. Lad was identified as a novel protein. As such, the method for selecting an adapter protein gene that binds to the SH region of the tyrosine kinase of the present invention is a Src family tyrosine kinase (p59 fyn , Blk, Yes, Lyn, Yrk, Hck, Fgr and Abl other than p56 lck ). Or Tec family tyrosine kinase may be useful for screening genes expressing adapter proteins that bind to SH2 regions. |
