abstract The present invention relates to the synthesis, separation and identification of N-demethylated and N, N-demethylated derivatives of two conformational isomers of toluidine blue O (“TBO”).n n n In vivo biocolored compositions include morphic isomers of TBO and their N- and N, N-demethylated derivatives. The ratio of these isomers to these demethylated derivatives is at least 6: 1.n n n Improved methods for detecting underdeveloped oral tissue include applying such TBO product to oral tissue with a liquid carrier.n n n The process for producing the TBO product comprises the steps of (1) oxidizing N, N-dimethyl-ρ-phenylenediamine with, for example, potassium dichromate to form 2-amino-5-dimethylaminophenyl thiosulfonic acid, (2) Oxidizing this intermediate, condensing the oxide with o-toluidine to form indaminthiosulfonic acid, and (3) further oxidizing the indamin intermediate to form a TBO containing reaction product, which precipitates from the reaction mixture as a complex. And separating the final TBO product therefrom. The complexing agent is added to the reaction mixture at least before the final oxidation step, preferably before the second oxidation step.n n n HPLC methods characterize TBO products containing form isomers of TBO and their N, N-demethylated derivatives. The mobile phase comprises an aqueous solution of an organic acid compound.
