titleOfInvention | Autoantibodies and Analysis Methods of Patients with Acquired Parathyroidism |
abstract | Acquired parathyroidism (AH) is a common component of type I autoimmune polyglandular syndrome (APSI) or occurs frequently as a sporadic isolated disease in adults. Autoantibodies to parathyroid glands have been reported in the AH, but their reality is still controversial. We report herein the detection of autoantibodies as confirmed through immunoblotting with serum of AH patients. Antibodies were specific for parathyroid autoantigens obtained by differential centrifugation of fresh human hypercellular parathyroid gland, obtained immediately after surgical removal for hyperparathyroidism in renal failure patients. Autoantibodies were detected for several parathyroid specific proteins in 17 AH patients tested. 12 (70%) reacted with 70 kDa protein, 16 (94%) with 80 kDa protein, and 3 (18%) with 120-140 kDa protein. 70 kDa and 80 kDa proteins were only present in the cytosol fraction of the parathyroid extract, and 120-140 kDa protein was present only in the membrane fraction. Antibodies can also be detected using the dog's parathyroid gland as an antigen source. However, antibodies could not be identified when using human melanocytes or rat pituitary cells cultured with antigen source. Serums from 50 other autoimmune patients and 22 normal controls were also tested, none of which reacted with the specific parathyroid proteins described above. It can be concluded that autoantibodies against three major autoantigens in human parathyroid extract were detected in AH patients. Moreover, our data indicate that 120-140 kDa autoantigens are calcium sensing receptors and reactive epitopes are mainly present in the extracellular domain. These data confirm the autoimmune properties of AH, and the present invention provides a method for detecting AH or AH as well as a method for treating AH. |
