abstract | The present invention provides a modified thermostable DNA polymerase with improved efficiency for conjugating non-conventional nucleotides such as those labeled with fluorescent dyes. Such modified thermostable DNA polymerases can be prepared by recombinant DNA techniques. The recombinant thermostable DNA polymerases produced according to the present invention are advantageous for many in vitro DNA synthesis applications such as DNA sequencing, synthesis of labeled DNA, and production of labeled primer extension products. The polymerase enzyme is particularly useful for the method of sequencing nucleic acid chains. Nucleic acids encoding the recombinant thermostable DNA polymerases of the invention as well as vectors and host cells comprising the same. A kit comprising the recombinant thermostable DNA polymerase of the invention is also provided. The polymerases and methods of the present invention provide significant cost and efficiency advantages over DNA sequencing. |
