abstract | The present invention relates to a recombinant expression vector using phosphoribulokinase as a fusion partner and a method for producing human parathyroid hormone using the same. More specifically, the present invention comprises a nucleotide sequence of a urokinase cleavage site in an L-arabinose inducible vector containing a phosphobulokinase gene fragment of Rhodobacter sphaeroides or a mutant thereof as a fusion partner. The present invention relates to a method for producing a large amount of human parathyroid hormone by culturing a recombinant expression vector prepared by inserting a gene of human parathyroid hormone, a recombinant microorganism transformed therefrom and an electric microorganism in a medium to which L-arabinose is added. Recombinant human PTH prepared in the present invention was confirmed to have the activity of native human PTH. Thus, by inducing the expression of L-arabinose by culturing the microorganism transformed with the recombinant expression vector of the present invention, the recombinant human PTH that maintains the activity of the native human PTH as it is produced in high yield through precise induction control can do. |
