abstract The present invention utilizes a cDNA encoding a novel protein (leukotactin-2: Lkn-2) belonging to C6 β-chemokine, isolated from a human mononuclear cell line (monocytic cell line), an expression vector comprising the same, an electric expression vector The present invention relates to a method for producing an electrical protein that attracts subsets of peripheral blood leukocytes. The open reading frame of Lkn-2 cDNA of the present invention encodes a total of 137 amino acids, including a signal peptide consisting of 21 amino acids, of which the molecular weight of the mature protein consisting of 116 amino acids is estimated to be 12,500 Daltons. Recombinant Lkn-2, purified by expressing the aforementioned Lkn-2 cDNA in recombinant E. coli or insect cells, significantly inhibits colony formation, has chemotaxis that attracts lymphocytes, monocytes and neutrophils, and hMIP-1α and RANTES It has been shown to bind to the receptor of binding CCR1. The recombinant Lkn-2 protein can be used for antibody production, treatment during HIV-1 infection, bone marrow hepatocyte protection during chemotherapy or radiation therapy, leukemia inhibitor, and the like.
