abstract The present invention provides primers used for next-generation sequencers capable of providing high read counts. N 5-15 is 5'-CAAGCAGAAGACGGCATACGAGAT-N 5-15 -GTCTCGTGGGCTCGGAGATGTGTATAAGAGACAG-3', representing an index sequence of 5 to 15 nucleotides. Based on the sequence, the index sequence is a nucleotide sequence representing the estimated number of reads exceeding a given level designed, and the estimated number of reads is calculated using an estimation formula specifying the number of reads as the objective variable and the nucleotide type of the index sequence as the explanatory variable.
