abstract | The present invention relates to the use of a novel compound for the prevention, improvement or treatment of amyotrophic lateral sclerosis, and the present inventors have found that SOD1 aggregation is one of the important causes in ALS disease, and suppression of regulation of TDP-43 or intracellular stress suggested the possibility that aggregation of WT-SOD1 by sALS could be the cause. In addition, the present inventors have discovered a new compound, PRG-A-01 (SLC-B036), as an inhibitor of SOD1 aggregation and misfolding. The compound showed a protective effect against muscle weakness and movement disorders in ALS model mice. As a result of histological analysis, the nerves in the spinal cord were maintained by treatment with PRG-A-01 (SLC-B036). In addition, the present inventors obtained a more optimized drugable candidate compound (PRG-A-04). In conclusion, the compound of the present invention can be usefully utilized for development as a therapeutic agent for ALS disease. |
