abstract |
Some aspects of the present disclosure provide strategies, methods, and reagents for determining nuclease target site preference and specificity of site-specific endonucleases. Some methods provided herein are invariant with candidate nuclease target sites to identify library members that could be cut by the nuclease of interest through sequencing of intact target sites that are identical to and adjacent to the cut target site. Utilizes a novel “one-cut” strategy for screening libraries of concatemers containing repeat units of insertion regions. Some aspects of the present disclosure provide strategies, methods, and reagents for selecting site-specific endonucleases based on determining their target site preference and specificity. Methods and reagents for determining target site preference and specificity are also provided. |
