abstract | The present invention relates to a single cell analysis method using magnetic beads, and more particularly, to a microwell-based single cell analysis method using magnetic streptavidin beads to which biotinylated cell-specific primers are bound. The single-cell analysis method (MAPS-seq) according to the present invention enables high-accuracy single-cell transcriptome analysis in multiple cells using magnetic beads coated with streptavidin to which biotinylated cell-specific primers are bound. In the initial stage of analysis, before the reverse transcription process, samples are pooled into one tube at once, so it is easy to perform the analysis without expensive equipment or advanced technical manipulation, and there is an effect of reducing the cost by reducing the waste of reagents. In addition, through specific verification experiments, it was confirmed that human and mouse cell lines can be effectively separated into various species by the above method, and that the method can be applied to drug response studies. The MAPS-seq method according to the present invention is improved convenience It is expected that it can be used for various research in related fields as a microwell-based assay with excellent cost-saving effect. |
