abstract The present invention relates to antisense oligonucleotide (ASO) compositions and in particular to compositions and methods for cytoplasmic delivery of antisense oligonucleotides (ASO). Hybrid ASOs are provided that are partially single-stranded and partially double-stranded, which hybridize to form a double-stranded region capable of non-covalently binding to a nucleic acid-binding protein region. In this way, ASO::protein complexes can be produced that facilitate delivery of antisense DNA into target cells. Such complexes can be used to downregulate gene expression in cells.
