abstract |
The present invention relates to an in vitro method for incorporating an exogenous DNA sequence into the genome of a cell using a transposon system. The transposon system comprises a first vector carrying an inverted repeat sequence and exogenous DNA, and a second vector expressing a transferase. Also provided is a kit comprising a transposon system for integrating exogenous DNA into the genome of a cell. A method of treating a subject is described, comprising the steps of engineering immune cells to retain an exogenous DNA sequence using the methods and/or kits described above, and administering to the subject an effective amount of the engineered immune cells. |
