abstract | The present invention provides an enzymatic method for measuring the concentration of one or more analytes in a plasma portion of a blood-derived sample containing a first component and a second component, wherein the second component is the measurement of the first component. Disturbs. The method comprises i) diluting the sample with a reagent mixture; ii) substantially removing blood cells; iii) using a reagent that temporarily inhibits the reaction of the second component to generate a blocked second component; iv) directly or indirectly generating detectable reaction products by inducing a selective reaction of constituents of each analyte, wherein one of the analytes is a first component; v) monitoring detectable reaction products or products; vi) correlating the detectable product or quantity of products and/or the rate of formation of the detectable product or products with the concentration of each analyte, wherein the concentration of at least the first component is not measurable It involves estimating the (imaginary) endpoint and associating it with a detectable corresponding reaction product. Step iii) may be performed at any step up to step iv) and including step iv), but before step v) or vi). The reagents of step iii) may be applied to the sample separately, or may be included in the reagent mixture during steps i) or iv). A corresponding kit is also provided. |
