titleOfInvention Foot-and-mouth disease virus-like particle vaccine and its manufacturing method
abstract The present invention provides a foot-and-mouth disease virus-like particle vaccine and a method for manufacturing the same. Constructing a recombinant expression plasmid by inserting two ORFs of the codon-optimized FMDV P12A and 3C genes into a transfer vector in series; Or, under the promoter SV40, a single ORF containing the P12A3C gene is cloned into a vector to construct a recombinant expression plasmid, transformed into competent cells to obtain recombinant Bacmid-DNA, and transfected into sf9 cells to obtain foot-and-mouth disease virus-like particle protein. Insect cells capable of expression were obtained. After culturing the cells, the supernatant was collected, and the VLP was purified by ultrafiltration concentration, centrifugation, and chromatography to prepare a foot and mouth virus-like particle vaccine. Foot-and-mouth disease virus-like particle vaccine of the present invention has excellent immunogenicity and safety, can induce the production of neutralizing antibodies of the animal itself, and can provide virus attack protection. Large-scale production and purification are possible using serum-free, protein-free medium and bioreactors and subsequent continuous chromatography purification.
