abstract | The present invention provides an enzymatic method for measuring the concentration of one or more analytes in the plasma portion of a blood derived sample containing a first component and a second component, wherein the second component is a measurement of the first component. Disturbs. The method comprises i) diluting the sample with a reagent mixture; ii) substantially removing blood cells; iii) using a reagent to temporarily inhibit the reaction of the second component to generate a blocked second component; iv) inducing a selective reaction of each analyte component to directly or indirectly generate detectable reaction products, wherein one of the analytes is a first component; v) monitoring the detectable reaction product or products; vi) correlating the amount of detectable product or products and / or the rate of formation of the detectable product or products with the concentration of each analyte, wherein the concentration of at least the first component is not measurable. Estimating the (virtual) endpoint and relating the corresponding reaction product that can be detected. Step iii) may be carried out in any step up to and including step iv) and before step v) or vi). The reagents of step iii) may be applied separately to the sample or may be included in the reagent mixture during step i) or iv). Corresponding kits are also provided. |
