titleOfInvention | Optogenetic protein clustering through fluorescent protein tagging and extension of CRY2 |
abstract | Homo-oligomerization of proteins is an important molecular mechanism in various biological fields. Thus the ability to modulate homo-oligomerization of proteins allows for the control and investigation of various cellular situations. To achieve this goal, Arabidopsis derived cryptochrome 2 (CRY2) was used to modulate the spatiotemporal protein homo-oligomerization by blue light. However, the exact mechanism of the CRY2 protein is not known, which hinders its widespread use. In the present invention, the inventors have found important elements for fast, effective CRY2 clustering, and have developed a system that can induce homo-oligomerization of fast and effective target proteins using engineered CRY2 proteins based on various fluorescent proteins and very short peptides. Developed. In addition, by comparing the light binding protein binding tools with the system of the present invention, it was confirmed that the system of the present invention can be used with a high efficiency in a much wider field. The present invention not only provides a novel light controlled protein clustering toolbox, but also provides guidelines for the development of a new optogenic module based on CRY-2. |
