abstract |
The present invention provides endoglycosidase referred to as EndoS49 and having the amino acid sequence of SEQ ID NO: 1. EndoS49 was isolated from Streptococcus pyogenes strain NZ131 and is homologous to EndoS. EndoS49 has endoglycosidase activity that is characteristic of natural IgG and is cleaved into more Fc glycans than EndoS. The mutant was substituted with glutamic acid at position 186 of SEQ ID NO: 1, and the mutant lacked endoglycosylase activity, but IgG binding is possible. Methods for using EndoS49, its deletion and methods for separating IgG, or evaluating the glycosylation of such variants, in particular IgG, are described. |
