abstract | The present invention relates to isolated laminin-521, a method for producing recombinant laminin-521, a host cell expressing recombinant laminin-521, and a composition containing laminin-521. Laminin-521 can maintain the pluripotency of stem cells in vitro, enable self-renewal, and enable single-cell survival of human embryonic stem cells. When pluripotent human embryonic stem cells are cultured on a plate coated with a substrate of recombinant laminin-521 (laminin 11), in the absence of differentiation inhibitor or supporting cells, the embryonic stem cells proliferate and retain their pluripotency. It has also been found that human recombinant laminin-521 (laminin-511) provides single cell survival of stem cells after complete dissociation into a single cell suspension. A useful cell culture medium containing up to 3.9 ng / ml of beta fibroblast growth factor (bFGF) was also described herein. |
