titleOfInvention | Method for producing sialic-acid-containing sugar chain |
abstract | [PROBLEMS] The importance of sialic acid? 2,3-linked or? 2,6-linked sugar chains at the non-reducing end of sugar chains is known, and industrial production of these sugar chain compounds is required. In particular, it is indispensable to mass-produce a sugar chain having a uniform structure by controlling the binding mode of sialic acid (α2,6 bond or α2,3 bond) in the production of a glycoprotein drug. It is considered that it is generally difficult to synthesize a sugar chain having sialic acid at all non-reducing ends chemically for the N-type complex sugar chain of the 3-branch type or the 4-branch type. It has been reported that the sugar chain is chemically synthesized none. Also, it has been difficult to efficiently prepare these sugar chains enzymatically. [Solution] In the present invention, it has been found that the sialic acid transferase has an activity of decomposing sialic acid on the reaction product in the presence of CMP newly, and enzymatically decomposes the produced CMP to thereby efficiently produce a sialic acid-containing sugar chain Respectively. In addition, the 4-branch type N-type sugar chain in which 4 molecules of sialic acid was added with α 2, 6 bond, which was difficult to synthesize conventionally, was also subjected to a sugar chain extension reaction using a bifurcated sugar chain as a raw material, Can be prepared at a high yield. |
