abstract The present invention relates to the detection of target nucleic acid sequences using PTO cleavage and extension-dependent signaling oligonucleotide hybridization (PCE-SH). The present invention does not use a probe that hybridizes with a target nucleic acid sequence to provide a target signal. The present invention utilizes probes (signaling oligonucleotides) that hybridize with extended strands formed in a target-dependent manner, and the extended strands are synthesized with a template CTO having an artificially selected sequence.
