abstract | The present invention relates to a method and composition for increasing the tricogenicity of a cell in a culture. One embodiment provides a method of increasing the tricogenicity of isolated mammalian epidermal cells compared to untreated isolated mammalian dermal cells by administering in vitro an effective amount of at least one Sonic hedgehog (Shh) pathway agonist Comprising culturing isolated mammalian dermal cells. The cell culture optionally contains epidermal cells. Preferred Shh agonists include, but are not limited to, commercially available CUR-0236715 and CUR-0201365 from Curitis in Corporation. Tricogenicity is measured using an Aderans Hair Patch Assay ( TM ). The cultured dermal cells may be maintained in the medium in the presence of one or more Shh agonists for at least one to seven days prior to harvest. Treated cultured dermal cells may be used to treat hair loss in a mammalian subject, preferably a human, by implanting in an amount effective to induce hair follicle formation. |
