abstract | There is provided a method for preparing a pharmaceutical composition containing a thermostable hemoglobin-based oxygen carrier favored by beta-beta crosslinking. The results of the present invention can be used to control the oxygen affinity of the resulting molecule and thus produce a hemoglobin-based oxygen carrier tailored to the particular application. Cross-linked hemoglobin with low oxygen affinity is useful for applications requiring rapid tissue oxygen supply (e.g., hemorrhagic shock), while cross-linked hemoglobin with high oxygen affinity requires low-rate oxygen supply Lt; / RTI > Highly purified and thermostable cross-linked non-polymeric cross-linked tetrameric hemoglobin is produced that has 40-60% or more of the beta-beta linkage and is suitable for use in mammals without causing renal damage. A high temperature short time (HTST) heat treatment step is performed to effectively remove undesirable dimeric forms of hemoglobin, noncrosslinked tetrameric hemoglobin, and plasma protein impurities. |
